| I. Background | |
信號(hào)轉(zhuǎn)導(dǎo)和轉(zhuǎn)錄激活蛋白（STAT）被確定為 DNA 結(jié)合蛋白的主要成分，該蛋白可響 應(yīng)多種細(xì)胞因子而激活基因轉(zhuǎn)錄。STAT 家族中的七個(gè)不同成員（STAT1，STAT2，STAT3， STAT4，STAT5A，STAT5B 和 STAT6）調(diào)節(jié)從病原體到細(xì)胞因子分泌的許多生理和病理過(guò) 程。STAT4 被多種細(xì)胞因子激活，包括 IL12、I 型干擾素（IFN-1）、IL23、IL2、IL27、 IL35 等。 | |
| II. Description | |
STAT4-Luc HEK293 報(bào)告基因細(xì)胞，是由 STAT4 調(diào)控并表達(dá) Luc 熒光素 酶報(bào)告基因的 HEK293 細(xì)胞。IFNγ與受體結(jié)合，激活 STAT4 的原理見(jiàn)下圖所示。
